Абстракт
Background: The production of reactive oxygen species (ROS) in mitochondria is very sensitive to the proton gradient and decreased by mild uncoupling, mediated e.g. by dinitrophenol. Rotenone is also a well characterized specific inhibitor ofcomplex I. In the study of placing the membrane - redox potential three state line systems enzyme substrate complex between donators - C6H12O6 and acceptors - O2 in the metabolic equation in the form as C6H12O6 + membrane - redox potentialthree state line systems enzyme substrate complex + 6O2 = energy + 6H2О +6CO2 and also revealing and correcting a omitted third (missing) member inchemical balance equation for the metabolism of every living cells we suppose thefollowing new model of functioning of human, animal body in accordance withsuch metabolic reaction as C6H12O6 + membrane - redox potential three state linesystems enzyme substrate complex + 6O2 = energy + 6H2О + 6CO2.
Objective: The aim of this study was to determine oxidase enzyme activity andcontents of deprotonized products in the second compartment during modeling of proton leak and proton block situation in rabbits.
Methods: The mitochondrial uncoupling was induced by dinitrophenol in theperiod of 3-56 days in rabbits. Subjects of our experiments were rabbits aged 18-24 months, including male and female Chinchilla rabbits, weighing 1800-2000 g.Induction in rabbits a high level of proton and electron conductance induced bysubcutaneous injection of 2,4-Dinitrophenol in dosage of 3.5mg/kg. Induction inrabbits a low level of heat energy release, high degree of energy accumulation andhigh degree of proceed ATP (adenosine-three-phosphate) induced by subcutaneousinjection of Rotenone in dosage of 2,5 mg/ kg for four days. In 3, 7, 14, 28, 56days was taken 4 ml of blood from ear’s marginal vein for analyses.
Results: Our results indicate that II compartment oxidase activity is increasedin dinitrophenol induced group, but decreased in rotenone induced group. Alsowe detected increasing of MDA in II compartment in proton leak constitution. Weobserved that in proton block MDA in II compartment was decreased.
Conclusions: In conclusions, results of our study show that in both Mkhrisand Rlung protons leaking is increasing, whereas in Badgan blockage of protonconductance is increasing.
